Subgroup analyses stratified by ethnicity and stage of DN were performed, and results indicated the Pro12Ala polymorphism was associated with the risk of DN in Caucasians but no similar association was observed in Asians.
The genetic influence of PPARG P12A genotype is modest and is overshadowed by duration of diabetes and systolic blood pressure as the major risk factors for diabetic nephropathy in the Oji-Cree population.
Our results support an important association between rs1805192 minor allele (G allele) of PPARG and increased DN risk; the interaction analysis showed a combined effect of interaction between rs1805192 and abdominal obesity on DN risk.
We conclude that P12A and C161T polymorphisms of the PPAR-gamma gene are important predictors of cardiovascular event in patients with diabetic nephropathy.
