Active vitamin D supplementation could potentially impede hepatocyte senescence and apoptosis via suppression of the p53 pathway, thus preventing the progression of NAFLD.
We used long-term pharmacological activation of p53 by i.p. or oral administration of low-dose doxorubicin in different animal models of NAFLD (high fat diet containing 45% and 60% kcal fat) and NASH (methionine- and choline-deficient diet and choline deficiency combined with high fat diet).
In this review, the crosstalk among p53, lipid metabolism, insulin resistance, inflammation and oxidative stress in NAFLD is discussed, and we suggest that a better understanding of p53 would present a promising potential new strategy for NAFLD prevention and treatment.
