We genotyped 7 SNPs (SG13S25, SG13S89, SG13S41, SG13S377, SG13S35, SG13S114, and SG13S32) in the ALOX5AP gene in 500 angiographically proven coronary artery disease (CAD) cases and 500 age- and gender-matched controls of European-American ancestry living in upper-Midwest US.
We also review in more detail studies that have revealed an association of ischemic stroke with polymorphisms of proinflammatory genes of particular interest (LTA, IL6, and ALOX5AP) as well as with polymorphisms at chromosomal region 9p21.3, which has recently been identified as a susceptibility locus for coronary heart disease.
ALOX5AP, a gene within the inflammatory leukotriene pathway linked to and associated with coronary atherosclerosis, is also associated with in-stent restenosis.
Studies that employ genome-wide linkage scans with hundreds of small nuclear families have identified new susceptibility genes for coronary artery disease and myocardiaI infarction, including ALOX5AP (encoding 5-lipoxygenase-activating protein) associated with myocardial infarction and stroke and PDE4D (encoding phosphodiesterase 4D) for ischemic stroke.
