In this excerpt from his forthcoming book on the history of cancer research, Joe Lipsick looks back at the discovery of tyrosine kinases and the demonstration that the V-SRC protein encoded by Rous sarcoma virus was a tyrosine kinase.

As described by Martin and Rous, a genetic recombination between c-src and the RSV oncogene of Rous sarcoma virus results in a modified Src protein, with increased intrinsic activity and transforming potential in animal and human tissues.

Activation of c-src, a cellular human gene homologous in sequence to the v-src gene of Rous sarcoma virus, had been thought to play an important role in the progression of several types of human cancers, without having undergone any genetic changes.

The antibody specifically reacted with N-myristoylated pp60c-src in human colon adenocarcinoma cells, N-myristoylated pp60v-src in Rous sarcoma virus-infected cells, and N-myristoylated Gag precursor protein Pr55gag in HIV-1 producing cells.

These results indicate that the human c-src gene has a potential transforming ability and suggest that the v-src-specific sequence played an important role in the generation of Rous sarcoma virus.

An endogenous protein of human cells pp60c-src, which is closely related to the product of the transforming gene of Rous sarcoma virus, pp60v-src, has been quantitated by measuring its enzymatic activity in an immunoglobulin G protein kinase assay.

The tyrosine protein kinase activity of the transformation gene product of Fujinami virus was particularly sensitive to the bromomethyl ketone derivative of leucine, while the src gene product of Rous sarcoma virus was comparatively less sensitive.

We have compared the polypeptide products of the sarc gene, p60(sarc), of human, mouse, and chicken cells with the polymorphic polypeptide product of the src gene, p60(src), of several strains of Rous sarcoma virus by two-dimensional peptide mapping. p60(sarc) from chicken cells was clearly related to every viral p60(src).
